513 Regulatory roles of lncRNAs in psoriasis determined by single cell analysis

Long noncoding RNAs (lncRNAs) can harbor DNA binding domains and regulate target gene expressions by to their promoter regions. Although a large proportion of differentially expressed lncRNAs have been identified in psoriatic skin, chronic inflammatory disease enriched pro-inflammatory genes, cell type specificity are yet be revealed. We performed scRNA-seq experiments non-lesional skin lesional from 6 psoriasis patients, compared the expression profiles against 5 control samples. Most up-regulated lncRNA were keratinocytes (for example, MALAT1 NEAT1), fibroblasts (MEG3), or melanocytes (KU-MEL-3). However, many these do not known functional roles. conducted silico analysis assay ability for 121 lesional/non-lesional (FDR<=5%) within each 12 types, revealing 5,879 potential downstream protein-coding targets. that NEAT1 (known perform regulatory roles) targets include epidermal genes such as: PPP1R15A, ZFP36, SFN, which responsible cycle regulation proliferation. Notably, all correlated with NEAT only but non-lesional/control keratinocytes. In addition, is also co-expressed other JUN, JUNB, involved response (R≥0.35, p<2.2e-16).
